GATE 2023 BM – Question 59
In a 24-hour cell-viability assay with 10000 cells and absorbance measured at 570 nm, control absorbance is 0.8, treated absorbance 0.5 and blank absorbance 0.1. Find percentage inhibition of viability to one decimal place.

Show answer and explanation
Correct answer: 42.0 to 44.0
Explanation
First remove the blank from both measurements: control net absorbance=.8−.1=.7, treated=.5−.1=.4. Relative viability=.4/.7, so inhibition=[1−.4/.7]×100=42.857%. Rounded to one decimal this is 42.9%. Using uncorrected readings would give the wrong inhibition.
