Viral thymidine kinase phosphorylates ganciclovir, initiating its conversion to a toxic nucleotide analogue. Cells retaining the viral tk marker therefore die under negative selection. Correctly targeted recombinants can be designed to lose that marker while keeping the positive-selection marker. Select B; the other kinases do not provide this selection mechanism.
